Introduction

1.What is vitamin B6?

Vitamin B6, also known as pyridoxine (Pyridoxine), is an important water-soluble B vitamin. It exists in the body in the forms of pyridoxine (PN), pyridoxal (PL), pyridoxine phosphate (PLP), pyridoxal phosphate (PNP). It participates in key biochemical processes such as amino acid metabolism and neurotransmitter synthesis.

Vitamin B6 is widely found in meat (fish and poultry), whole grains, vegetables and nuts. As an essential micronutrient for maintaining human health, growth and normal physiological functions, it possesses various physiological functions including anti-inflammatory, antioxidant, neuroregulatory and antitumor effects. It plays a crucial role in maintaining nervous system health, skin health and immune function.

2.How it Works?

Vitamin B6 (pyridoxine) is a water-soluble vitamin whose core mechanism of action involves serving as a coenzyme in over 100 enzymatic reactions, particularly in processes such as amino acid metabolism, neurotransmitter synthesis, and heme production. Its active form, pyridoxal phosphate (PLP), regulates protein, fat, and carbohydrate metabolism through binding with enzymes, while directly influencing nervous system function, immune system performance, and gene expression.

3.Benefits and Use

Metabolic Functions
Vitamin B6 is essential for maintaining normal growth, development, and cellular metabolism. Pyridoxal phosphate (PLP), the biologically active form of vitamin B6, serves as a coenzyme factor in over 140 biochemical reactions in cells. These include critical metabolic processes such as the synthesis, interconversion, and degradation of amino acids; the metabolism of carbohydrates, lipids, and nucleic acids; and heme synthesis. Deficiency in vitamin B6 disrupts normal protein and amino acid metabolism, reduces protein absorption and utilization efficiency, and ultimately leads to symptoms such as anorexia, growth retardation, neurological disorders, and anemia in animals.

Neuromodulatory Functions
Vitamin B6 acts as a cofactor in the synthesis of neurotransmitters, including epinephrine, dopamine, serotonin (5-hydroxytryptamine, 5-HT), and γ-aminobutyric acid (GABA), playing a vital role in the normal development and function of the nervous system. Serotonin regulates cognition, mood, and sleep and is commonly targeted by sedatives, antidepressants, and anxiolytics. GABA is an inhibitory neurotransmitter with calming effects. By modulating neurotransmitter synthesis, vitamin B6 influences mental health and cognitive function, exhibiting neuroprotective properties. Deficiency in vitamin B6 is associated with various neurological disorders.

The ATP-gated purinergic P2X7 receptor (P2X7) acts as an inflammatory switch in the brain, mediating the release of fast excitatory neurotransmitters in the central and peripheral nervous systems. It promotes the proliferation of astrocytes and microglia, increases the release of pro-inflammatory cytokines and other inflammatory mediators, and triggers epileptic seizures. P2X7 is considered a novel drug target for treating neurological disorders, including epilepsy. Studies have shown that vitamin B6 acts as a P2X7 receptor antagonist, exhibiting antiepileptic and neuroprotective effects.

Antioxidant Functions
The pyridine ring of vitamin B6 contains hydroxyl and amino groups, enabling it to directly react with reactive oxygen species (ROS) and function as an effective antioxidant. Additionally, vitamin B6 participates in the production of various bioactive compounds. Both taurine and hydrogen sulfide (H₂S) are derived from sulfur-containing amino acids, and their synthesis requires vitamin B6-dependent enzymes.

Immunomodulatory and Anti-inflammatory Functions
Vitamin B6 is a critical compound for maintaining innate and adaptive immune system functions, particularly in anti-inflammatory immune responses. Research indicates that vitamin B6 deficiency impairs cellular and humoral immune responses, leading to weakened immunity. Manifestations include atrophy of lymph nodes, lymphoid organs, and the spleen; reduced lymphocyte counts; decreased production of antibodies (immunoglobulin G [IgG] and immunoglobulin M [IgM]); and altered cytokine production.

Antitumor Functions
Epidemiological studies have reported an inverse correlation between vitamin B6 intake and the risk of colon cancer. Research suggests that the antitumor effects of vitamin B6 are primarily linked to the inhibition of cell proliferation, oxidative stress, inflammatory responses, nitric oxide (NO) synthesis, and angiogenesis. Komatsu et al. found that supplementing vitamin B6 in the diet of colorectal cancer mice significantly reduced tumor incidence and the number of colon tumors while suppressing cell proliferation. It also markedly decreased the protein expression of proto-oncogenes (c-Myc and c-Fos) associated with cell proliferation in colonic crypts, without significantly affecting apoptosis in colon epithelial cells. In vitro studies have demonstrated that vitamin B6 inhibits the proliferation and migration of monocytic lymphoma cells, increases IL-8 and IL-10 levels, reduces IL-1β production, and impedes tumor progression.
